AMP-activated protein kinase (AMPK) is the central energy sensor of eukaryotic cells, a heterotrimeric serine/threonine kinase that monitors the cellular AMP:ATP and ADP:ATP ratios to restore energy homeostasis. When energy charge falls, adenine nucleotides bind the regulatory gamma subunit and promote phosphorylation of a conserved threonine in the activation loop of the catalytic AMPK-alpha subunit. The principal upstream kinase responsible for this modification is the tumor suppressor LKB1/STK11, which acts in complex with STRAD and MO25 to constitutively prime AMPK for activation under stress. The resulting Phospho-AMPK-alpha species marks the active enzyme and is a widely used readout of metabolic stress. Once engaged, AMPK switches off anabolic, ATP-consuming pathways and switches on catabolic, ATP-generating ones. It directly phosphorylates and inhibits acetyl-CoA carboxylase (ACC), the committed step of fatty acid synthesis, thereby shifting cells toward fatty acid oxidation. AMPK also restrains the mechanistic target of rapamycin complex 1 by phosphorylating TSC2, enhancing its GAP activity toward Rheb, and by directly phosphorylating Raptor, a core mTORC1 scaffold, which suppresses protein synthesis and growth. In parallel, AMPK promotes autophagy through activating phosphorylation of ULK1, including the characteristic Phospho-ULK1-S555 mark, coupling nutrient scarcity to the recycling of cellular components. Because this network governs the balance between growth and survival, its dysregulation contributes to type 2 diabetes, obesity, cardiovascular disease, and cancer, and AMPK is the molecular target of the widely prescribed antidiabetic agent metformin. Studying LKB1, AMPK-alpha and its phosphorylation, ACC, TSC2, Raptor, and the ULK1 axis together provides a comprehensive view of how cells perceive and respond to energetic stress.
