The intrinsic apoptotic pathway is governed by the balance of pro- and anti-apoptotic BCL-2 family proteins at the mitochondrial membrane. Pro-apoptotic signals shift this balance toward BAX/BAK oligomerisation, mitochondrial outer membrane permeabilisation and cytochrome c release, triggering APAF-1/caspase-9 apoptosome assembly and downstream caspase-3 activation.
