The construction of neural circuits depends on the precise navigation of growing axons and the dynamic remodeling of the neuronal cytoskeleton. Growth cones at the tips of extending axons integrate attractive and repulsive cues to steer toward their targets, and this guidance machinery is also reactivated during regeneration and plasticity. Several classic guidance systems are represented here. Semaphorin 3A is a secreted repulsive cue that collapses growth cones, and it signals through a receptor complex in which Neuropilin-1 / NRP1 serves as the principal ligand-binding subunit. Slit-Robo signaling provides midline repulsion, with Robo1 acting as the receptor that prevents inappropriate crossing of axons. The Eph-ephrin system contributes contact-mediated repulsion and topographic mapping, exemplified by the receptor EphA4, which regulates axon pathfinding and synaptic organization. The structural and motile properties of axons rely on a coordinated cytoskeleton. Neurofilament (NF-L), the light subunit of neuronal intermediate filaments, provides axonal caliber and structural stability and is a sensitive marker of axonal injury and degeneration. Tau stabilizes axonal microtubules and regulates their assembly and transport. Two markers track growth and structural plasticity directly: GAP-43 is a growth-associated phosphoprotein enriched in growth cones and regenerating axons that reflects active outgrowth, while Doublecortin/DCX is a microtubule-binding protein expressed in migrating neuroblasts and immature neurons, serving as a definitive marker of neurogenesis and neuronal migration. Together these targets connect the extracellular guidance cues that direct wiring to the intracellular cytoskeletal proteins that execute movement and maintain axonal integrity, processes whose disruption underlies developmental malformations, axonopathies, and failed regeneration after injury.
