The RAS-RAF-MEK-ERK cascade is a canonical mitogen-activated protein kinase module that converts extracellular growth and differentiation cues received at receptor tyrosine kinases into changes in gene expression, proliferation, and survival. Active GTP-bound RAS recruits and activates RAF kinases, including Raf-1 and B-Raf, at the plasma membrane. These activated RAF proteins phosphorylate and switch on the dual-specificity kinases MEK1/2, a regulatory event detectable as Phospho-MEK1/2. Activated MEK in turn phosphorylates the terminal kinases ERK1/2 on their threonine and tyrosine residues, and the resulting Phospho-ERK1/2 serves as the principal readout of pathway flux. Once active, ERK1/2 dimerize and translocate to the nucleus, where they phosphorylate substrates that drive a mitogenic transcriptional program. Among these, the ribosomal S6 kinase RSK is activated to regulate translation and cell-cycle entry, while the ETS-family transcription factor Elk-1 is phosphorylated to cooperate with serum response factor in inducing immediate-early genes. This pathway is among the most frequently deregulated in human cancer. Activating mutations in upstream RAS, and in B-Raf most notably the V600E substitution, lock the cascade in a constitutively active state, driving uncontrolled proliferation in melanoma, colorectal, thyroid, and other malignancies. Such addiction has yielded clinically important RAF and MEK inhibitors, although adaptive feedback reactivation and paradoxical RAF dimerization complicate sustained inhibition. Measuring both total and phosphorylated forms of MEK1/2 and ERK1/2 alongside the activators Raf-1 and B-Raf and the effectors RSK and Elk-1 provides a quantitative picture of cascade activity and inhibitor response.
