Upon activation, naive CD4 T cells differentiate into functionally specialized helper subsets whose identities are dictated by lineage-defining master transcription factors acting downstream of cytokine-driven STAT signals. The cytokine milieu sensed at priming activates particular STAT proteins that induce the corresponding master regulator, which then reinforces its own lineage while repressing alternatives. The Th1 program, geared toward intracellular pathogens, is initiated when IL-12 activates STAT4, leading to a transcriptional circuit that includes Runx3, a factor that cooperates to enforce cytotoxic and Th1 gene expression and to silence the opposing Th2 program. The Th2 lineage, central to antiparasitic and allergic responses, is governed by GATA3, the master Th2 factor induced by IL-4-driven STAT6 signaling, which drives the type 2 cytokine locus. The Th17 lineage, important in mucosal defense and autoimmunity, depends on the nuclear receptor RORgammaT, which programs IL-17 production. Regulatory T cells, essential for immune tolerance and suppression of excessive inflammation, are defined by the forkhead transcription factor FoxP3, the lineage-specifying and stabilizing regulator of suppressive function. Follicular helper T cells that support germinal center responses and antibody maturation are orchestrated by the transcriptional repressor BCL6. The multifunctional factor IRF4 acts across several effector lineages, dose-dependently shaping Th2, Th17, follicular helper, and regulatory differentiation in concert with these master regulators. The balance among these programs determines the quality of immunity, and its disruption underlies autoimmunity, allergy, chronic inflammation, and immune evasion. Defining subset identity in tissue and culture requires reagents recognizing each master transcription factor.
