The Ethanol Test Kit (SKU: BA0039) provides a simple, direct and automation-ready method for measuring ethanol concentration. Quantitative determination of alcohol (ethanol) finds applications in basic research, drug discovery, clinical studies and the wine industry, where heavy consumption is associated with liver disease and increased mortality. This kit is based on an improved dichromate method in which dichromate is reduced by ethanol to a bluish chromic (Cr3+) product; the intensity measured at 580 nm is a direct measure of the alcohol concentration in the sample. The optimised formulation substantially reduces interference from substances in raw samples and offers high sensitivity across a detection range of 0.04 to 2% alcohol. The procedure involves adding a single working reagent, incubating, adding a stop reagent and reading the optical density, and is readily automated for high-throughput work. Both 96-well plate and cuvette protocols are provided.

A colorimetric assay for the determination of ethanol. Based on an improved dichromate method, dichromate is reduced by ethanol to a bluish chromic (Cr3+) product; the intensity measured at 580 nm is a direct measure of the alcohol concentration. Detection range is 0.04 - 2% alcohol.

| 1 | Prepare 600 µL 2% Premix by mixing 120 µL 10% Standard and 480 µL distilled water, then dilute as shown in the standards table. Transfer 100 µL standards and samples into wells of a clear-bottom 96-well plate. |
| 2 | Add 100 µL Reagent A quickly using a multi-channel pipettor and tap lightly to mix. |
| 3 | Incubate 8 to 30 min at room temperature; the colour changes from yellow to bluish in the higher standards. Add 100 µL Stop Reagent B quickly and tap to mix. |
| 4 | Read OD at 570-600 nm (peak 580 nm). |
| 5 | Cuvette format: prepare 2%, 1% and 0.5% standards with water as blank, transfer 400 µL diluted Standards and samples to tubes, add 400 µL Reagent A, incubate 8-30 min, add 400 µL Reagent B, then read at 570-600 nm. |
Subtract the blank OD (water, standard #8) from the standard OD values and plot OD against alcohol concentration. Determine the sample ethanol concentration from the standard curve. Conversions: 1% (v/v) ethanol equals 170 mM or 785 mg/dL.
