Chronic antigen exposure drives T-cell exhaustion characterised by sustained expression of inhibitory checkpoints PD-1, CTLA-4, LAG-3, TIM-3 and TIGIT and the transcription factors TOX and EOMES. These molecules are central targets of cancer immunotherapy.
