The cellular heterogeneity of nervous tissue requires a panel of lineage- and maturation-specific markers to distinguish neurons, glia, and their progenitors in histological and molecular studies. Mature neurons are most commonly identified by NeuN/RBFOX3, a neuron-specific nuclear RNA-binding protein expressed throughout the differentiated neuronal population, and by NSE / ENO2, the neuron-specific enolase isozyme that reflects neuronal metabolic identity. The dendritic compartment of neurons is delineated by MAP2, a microtubule-associated protein enriched in cell bodies and dendrites that is widely used to visualize neuronal morphology and to distinguish dendrites from axons. Glial populations are resolved by complementary markers. GFAP labels astrocytes through its intermediate filament network, while S100beta is a calcium-binding protein expressed in astrocytes and other glia that marks mature astrocytic cells. The oligodendrocyte lineage is captured by Olig2, a basic helix-loop-helix transcription factor that specifies and maintains oligodendrocyte identity across progenitor and mature stages. Oligodendrocyte precursor cells, which persist throughout the adult brain and can proliferate in response to injury, are identified by NG2/CSPG4, a chondroitin sulfate proteoglycan that marks this dividing glial population. Neural stem and progenitor cells, present during development and in adult neurogenic niches, express Nestin, an intermediate filament protein characteristic of undifferentiated and reactive cells. Collectively these markers allow accurate assignment of cell type, assessment of differentiation state, and quantification of population shifts that accompany development, injury, and disease, providing the foundational framework on which more specialized analyses depend.
