The Glucose Assay Kit (Colorimetric) (SKU: BA0041) offers a simple, direct and automation-ready method for the quantitative colorimetric determination of glucose in serum, plasma, food and beverages. Glucose is a ubiquitous fuel molecule, and its concentration is a key diagnostic parameter for many metabolic disorders, with raised levels associated with diabetes mellitus and hyperactivity of the thyroid, pituitary and adrenal glands, and reduced levels found in insulin-secreting tumours, myxoedema and hypopituitarism. The assay measures glucose directly in serum or plasma without pretreatment using an improved o-toluidine method that produces a specific colour reaction with glucose. The absorbance at 630nm is directly proportional to the glucose concentration in the sample. As little as 5 µL of sample is required, making the kit both sensitive and economical. The optimised formulation greatly enhances reagent and signal stability.

This kit measures glucose directly in serum or plasma without pretreatment. The improved o-toluidine method utilises a specific colour reaction with glucose, and the absorbance at 630nm is directly proportional to glucose concentration in the sample.

| 1 | Dilute standard in distilled water as shown in the table. Set up 1.5-mL centrifuge tubes and transfer 5 µL diluted standards and samples to labelled tubes. Add 500 µL Reagent to each tube, close tightly and mix. Diluted standards may be stored at -20°C for future use. |
| 2 | Place tubes in a holder and heat in a boiling water bath or heat block for 8 minutes, then cool in a cold water bath for 4 minutes. |
| 3 | Transfer 200 µL in duplicate into a clear bottom 96-well plate, avoiding bubble formation, and read optical density at 620-650nm (peak absorbance at 630nm). |
Subtract blank OD (water, #5) from the standard OD values and plot OD against standard concentrations. Determine the slope by linear regression. Glucose concentration = (ODSAMPLE − ODBLANK) / Slope (mg/dL). Typical serum/plasma glucose values: 70-110 mg/dL. Conversion: 1 mg/dL glucose equals 55.5 µM, 0.001% or 10 ppm.
