The Protein Creatinine Ratio Assay Kit (SKU: BA0055) provides a simple, sensitive and automation-ready method for determining the protein creatinine ratio in urine samples. Protein is filtered out of urine by the glomeruli of the kidneys, and a damaged kidney allows some protein through, with elevated urinary protein (microalbuminuria or proteinuria) typically arising from type 1 or type 2 diabetes or high blood pressure. Creatinine is synthesised at a fairly constant rate, so normalising protein concentration to creatinine gives the protein creatinine ratio, which remains the simplest and most convenient test for proteinuria and allows a spot urine sample to be taken at any time without diet or liquid restrictions. The kit requires only 20 µL of sample and no pretreatment, using a simple 10-minute add-incubate-read procedure for each determination. The procedure can be readily automated for processing thousands of samples per day.

This kit determines the protein creatinine ratio in urine. Protein is measured at 600nm and creatinine at 530nm using internal standards, and the ratio of the two normalises protein excretion to creatinine secretion.

| 1 | Equilibrate all components to room temperature. Protein determination: transfer 20 µL of each sample into four wells (two Sample, two Internal Standard). Add 5 µL dH2O to Sample wells and 5 µL Standard to Internal Standard wells. Transfer 25 µL dH2O into two Blank wells. |
| 2 | Add 200 µL PR Reagent to each protein determination well, incubate 10 minutes at room temperature and read OD at 600nm. |
| 3 | Creatinine determination: set up sample, internal standard and blank wells as above. Prepare Working Reagent (per well: 50 µL CR Reagent A, 50 µL CR Reagent B, 150 µL dH2O) and transfer 200 µL into each creatinine well. |
| 4 | Incubate 10 minutes at room temperature and read OD at 530nm. |
Protein concentration = [(ODSTANDARD − ODSAMPLE) / (ODSAMPLE − ODBLANK)] × 10000 × n (µg/dL). Creatinine concentration = [(ODSTANDARD − ODSAMPLE) / (ODSAMPLE − ODBLANK)] × 25 × n (mg/dL). Protein Creatinine Ratio = [Creatinine] / [Protein] (µg/mg). A ratio below 30 is considered normal, 30-300 mild proteinuria, and above 300 severe proteinuria.
