The Hippo pathway is a conserved kinase cascade that controls organ size, tissue regeneration, and contact inhibition by restraining the activity of the transcriptional coactivators YAP and TAZ. When the pathway is active, the upstream kinase MST1, together with its paralog and the adaptor scaffold, phosphorylates and activates the kinase LATS1, a step facilitated by the adaptor MOB1. Activated LATS1 then phosphorylates YAP, generating the cytoplasmic Phospho-YAP form, and similarly phosphorylates TAZ; this modification creates binding sites for 14-3-3 proteins that sequester YAP and TAZ in the cytoplasm and promote their degradation, thereby keeping the pathway output off. The pathway is regulated upstream by cell-cell contact, mechanical cues, and cytoskeletal tension, with the tumor suppressor Merlin/NF2 acting as a key activator of the core kinase cassette in response to adhesion signals. When Hippo signaling is low, dephosphorylated YAP and TAZ accumulate in the nucleus, where they bind the TEAD family of transcription factors, exemplified by TEAD1, to drive expression of genes promoting proliferation, survival, and stemness. In cancer, this pathway is frequently disabled: loss of Merlin/NF2 or core kinase components, and amplification or nuclear enrichment of YAP and TAZ, sustain unrestrained transcriptional output that fuels tumor growth, metastasis, and resistance to targeted therapy. Disrupting the YAP/TAZ-TEAD interaction is an emerging therapeutic strategy. Measuring total and phosphorylated YAP, TAZ levels, the kinases LATS1 and MST1, the adaptor MOB1, the effector TEAD1, and the upstream regulator Merlin/NF2 resolves the activation state of the pathway.
