FGF21 Fibroblast growth factor 21

FGF21 (fibroblast growth factor 21) is a secreted endocrine member of the fibroblast growth factor family that acts as a major metabolic regulator. It stimulates glucose uptake in adipose tissue and helps regulate systemic glucose homeostasis and insulin sensitivity. Target information

What is FGF21 (fibroblast growth factor 21)?

FGF21 is a secreted protein that works as an endocrine factor and is described as a major metabolic regulator. It stimulates glucose uptake in differentiated adipocytes by inducing expression of the glucose transporter SLC2A1 (GLUT1). It does not induce SLC2A4 (GLUT4) expression. This activity requires the presence of KLB. Through these actions, FGF21 contributes to the regulation of systemic glucose homeostasis and insulin sensitivity, which makes it a frequent subject in metabolic research.

The FGF21 gene encodes a member of the fibroblast growth factor (FGF) family. FGF family members have broad mitogenic and cell survival activities and take part in many biological processes. UniProt places FGF21 in the heparin-binding growth factors family. The mature protein is secreted rather than retained in the cell, in keeping with its endocrine role. The mouse ortholog is Fgf21, so the same biology can be studied in murine models. Researchers use the FGF21 protein, FGF21 antibody and FGF21 ELISA kit formats to study its signalling and circulating levels.

HGNC classifies the FGF21 gene within the fibroblast growth factor family. Its endocrine mode of action, and its dependence on KLB, set it apart from many other family members that act locally as mitogens. This makes FGF21 a useful reference point when comparing metabolic and paracrine FGF signalling.

Glucose homeostasis research

FGF21 regulates systemic glucose homeostasis, so it is studied in work on how glucose handling is controlled across tissues.

Insulin sensitivity studies

FGF21 is reported to regulate insulin sensitivity, making it relevant to research on metabolic signalling and insulin response.

Adipocyte biology

FGF21 stimulates glucose uptake in differentiated adipocytes by inducing SLC2A1/GLUT1 expression, a mechanism examined in adipose tissue studies.

Endocrine FGF signalling

As a secreted endocrine factor whose activity requires KLB, FGF21 is a model for studying hormone-like FGF signalling.

Background

FGF21 frequently asked questions

What does FGF21 do?

FGF21 is a secreted endocrine factor and major metabolic regulator. It stimulates glucose uptake in differentiated adipocytes by inducing SLC2A1/GLUT1 expression, but not SLC2A4/GLUT4. It also regulates systemic glucose homeostasis and insulin sensitivity. Its activity requires the presence of KLB.

Which protein family does FGF21 belong to?

FGF21 is a member of the fibroblast growth factor family, which UniProt places in the heparin-binding growth factors family. Family members have broad mitogenic and cell survival activities. FGF21 is distinctive because it acts as a secreted endocrine factor in metabolism.

Is FGF21 conserved in mouse?

Yes, the mouse ortholog is Fgf21, so the gene can be studied in murine systems. No rat ortholog is given in the data supplied here. As always, check the datasheet of any product for its validated reactivity.
