P-Selectin Selectin P · granule membrane protein 140 (GMP-140, CD62P)

P-Selectin (selectin P, also known as CD62P, PADGEM, GMP140 and PSEL) is a calcium-dependent cell adhesion receptor of the selectin family. It mediates the interaction of activated endothelial cells or platelets with leukocytes during the early stages of inflammation. Target information

What is P-Selectin (selectin P)?

P-Selectin is a Ca(2+)-dependent receptor for myeloid cells. It binds carbohydrates on neutrophils and monocytes, and the ligand it recognises is sialyl-Lewis X. By linking activated endothelial cells or platelets to leukocytes, it supports the rapid rolling of leukocytes over vascular surfaces, which is one of the initial steps of inflammation. This rolling depends on its interaction with P-selectin glycoprotein ligand-1 (SELPLG). P-Selectin also mediates cell-cell interactions and cell adhesion through integrin alpha-IIb/beta3 (ITGA2B:ITGB3) and integrin alpha-V/beta-3 (ITGAV:ITGB3).

The SELP gene encodes a membrane protein that is stored in the alpha-granules of platelets and in the Weibel-Palade bodies of endothelial cells. When platelets are activated and degranulate, the protein redistributes to the plasma membrane, where it can engage sialylated forms of Lewis blood group carbohydrate antigens on neutrophils and monocytes. Alternative splice variants may occur but are not well documented. UniProt places the P-Selectin protein in the selectin/LECAM family, and HGNC lists it among the CD molecules, selectins, sushi domain containing and C-type lectin domain containing groups.

The protein has been known under several names, reflecting how it was first identified: granule membrane protein 140 (GMP-140), PADGEM, CD62 and CD62P. HGNC previously named the gene selectin P (granule membrane protein 140kDa, antigen CD62). Today the approved symbol is SELP, and the name P-Selectin is used widely for the protein in research on adhesion, platelet biology and leukocyte recruitment.

Leukocyte rolling and inflammation

P-Selectin mediates the rapid rolling of leukocytes over vascular surfaces in the initial steps of inflammation, through its interaction with SELPLG.

Platelet activation studies

The protein is stored in platelet alpha-granules and moves to the plasma membrane on activation and degranulation, making it a reference for platelet responses.

Endothelial cell biology

P-Selectin is held in endothelial Weibel-Palade bodies and mediates the interaction of activated endothelial cells with leukocytes.

Integrin-mediated cell adhesion

P-Selectin supports cell-cell interactions and adhesion through integrin alpha-IIb/beta3 and integrin alpha-V/beta-3.

Background

P-Selectin frequently asked questions

What does P-Selectin do?

P-Selectin is a calcium-dependent receptor that binds sialyl-Lewis X and related carbohydrates on neutrophils and monocytes. It mediates the interaction of activated endothelial cells or platelets with leukocytes and supports leukocyte rolling along vessel walls early in inflammation, via SELPLG.

What other names is P-Selectin known by?

The gene symbol is SELP and the HGNC name is selectin P. Aliases include CD62, CD62P, PSEL, PADGEM, GMP140, GMP-140 and GRMP. Older names refer to granule membrane protein 140 and antigen CD62. These all describe the same protein, so literature searches should cover several terms.

Where is P-Selectin found in the cell and does it have orthologs?

P-Selectin is stored in platelet alpha-granules and endothelial Weibel-Palade bodies, and it moves to the plasma membrane when platelets are activated. The rat ortholog is listed as Selp. Check the datasheet of any product for its validated reactivity.
