TUBGCP4 Gamma-tubulin complex component 4 · GCP4

TUBGCP4 (tubulin gamma complex component 4, also known as GCP4, 76P and FLJ14797) is a component of the gamma-tubulin ring complex (gTuRC), which mediates microtubule nucleation. The protein localises to centrosomes in association with gamma-tubulin. Browse Assay Genie's current range of TUBGCP4 research reagents below, or read on for the biology behind the target.

Target information

What is TUBGCP4 (tubulin gamma complex component 4)?

TUBGCP4 is a component of the gamma-tubulin ring complex (gTuRC), the assembly that mediates microtubule nucleation. The gTuRC regulates minus-end nucleation of alpha-beta tubulin heterodimers, which then grow into microtubule protofilaments. This is a critical step in centrosome duplication and spindle formation, so the TUBGCP4 protein sits at the base of microtubule organisation in dividing cells.

In mammalian cells, TUBGCP4 localises to centrosomes in association with gamma-tubulin. Crystal structure analysis revealed a fold composed of five helical bundles arranged around conserved hydrophobic cores. An exposed surface area in the C-terminal domain is essential and sufficient for direct binding to gamma-tubulin, which explains how the subunit is anchored within the complex. The protein belongs to the TUBGCP family, and HGNC classifies the gene within the tubulin gamma complex component family. The TUBGCP4 gene also undergoes alternative splicing, producing multiple transcript variants.

TUBGCP4 was previously named tubulin, gamma complex associated protein 4, and is also referred to as GCP4. Mutations in the gene that alter microtubule organisation are associated with microcephaly and chorioretinopathy, linking gTuRC integrity to neurodevelopmental and retinal phenotypes. The mouse ortholog is designated Tubgcp4.

Microtubule nucleation

TUBGCP4 is a gTuRC component that mediates minus-end nucleation of alpha-beta tubulin heterodimers into microtubule protofilaments.

Centrosome and spindle biology

The gTuRC contributes to centrosome duplication and spindle formation, and TUBGCP4 localises to centrosomes alongside gamma-tubulin.

Structural biology

Crystal structure work shows five helical bundles and a C-terminal surface that directly binds gamma-tubulin, making TUBGCP4 a model for complex assembly.

Neurodevelopmental disease

Mutations that alter microtubule organisation are associated with microcephaly and chorioretinopathy, making TUBGCP4 relevant to developmental disorder research.

Background

TUBGCP4 frequently asked questions

What does TUBGCP4 do?

TUBGCP4 is a component of the gamma-tubulin ring complex, which mediates microtubule nucleation. The complex regulates minus-end nucleation of alpha-beta tubulin heterodimers that grow into protofilaments, a critical step in centrosome duplication and spindle formation. TUBGCP4 binds gamma-tubulin directly through its C-terminal domain.

What other names and isoforms does TUBGCP4 have?

TUBGCP4 is also known as GCP4, 76P and FLJ14797. Its full name is tubulin gamma complex component 4, previously tubulin, gamma complex associated protein 4. UniProt names the protein gamma-tubulin complex component 4. Alternative splicing of the gene results in multiple transcript variants.

Is TUBGCP4 linked to disease or conserved across species?

Mutations in TUBGCP4 that alter microtubule organisation are associated with microcephaly and chorioretinopathy. The mouse ortholog is Tubgcp4. Researchers should check the datasheet of any product for its validated reactivity before use in a given species.
