VANGL2 (VANGL planar cell polarity protein 2, also known as Vang-like protein 2, KIAA1215, LTAP, LPP1 and STBM) is a cell membrane protein of the Vang family. It regulates planar cell polarity and contributes to early morphogenesis and neural plate development. What is VANGL2 (VANGL planar cell polarity protein 2)?

VANGL2 is a membrane protein involved in the regulation of planar cell polarity, the coordinated orientation of cells within the plane of a tissue. The encoded protein transmits directional signals to individual cells or groups of cells in epithelial sheets. It is best characterised in the cochlea, where it plays a role in orienting the stereociliary bundles of sensory hair cells.

Beyond the inner ear, VANGL2 is involved in the control of early morphogenesis and in the patterning of axial midline structures, as well as in development of the neural plate. It is required for the polarisation and movement of myocardializing cells in the outflow tract, and appears to act via RHOA signalling to regulate this process. VANGL2 is also required for cell surface localisation of FZD3 and FZD6 in the inner ear, linking it to Frizzled receptor biology.

The VANGL2 gene encodes a protein that belongs to the Vang family and localises to the cell membrane. The name reflects its relationship to Van Gogh in Drosophila: earlier HGNC names for the gene were vang (van gogh, Drosophila)-like 2 and vang-like 2 (van gogh, Drosophila). Other aliases in the literature include KIAA1215, LTAP, LPP1, STBM, STB1 and STBM1. Orthologues are annotated in mouse and rat, both named Vangl2, which allows polarity and developmental questions to be studied in these species.

Planar cell polarity research

VANGL2 regulates planar cell polarity and transmits directional signals across epithelial sheets, making it a central protein for studying tissue organisation.

Inner ear development

VANGL2 orients stereociliary bundles in the cochlea and is required for cell surface localisation of FZD3 and FZD6 in the inner ear.

Neural plate morphogenesis

VANGL2 is involved in early morphogenesis, patterning of axial midline structures and development of the neural plate.

Cardiac outflow tract development

VANGL2 is required for polarisation and movement of myocardializing cells in the outflow tract, apparently acting via RHOA signalling.

Background

VANGL2 frequently asked questions

What does VANGL2 do?

VANGL2 regulates planar cell polarity, passing directional signals between cells in epithelial sheets. It orients stereociliary bundles in the cochlea, contributes to neural plate and axial midline development, and supports myocardializing cell movement in the outflow tract, apparently via RHOA signalling.

What other names is VANGL2 known by?

The HGNC name is VANGL planar cell polarity protein 2, and UniProt lists Vang-like protein 2. Aliases include KIAA1215, LTAP, LPP1, STBM, STB1, STBM1, MGC119403 and MGC119404. Earlier names referred to the Drosophila gene van gogh, as in vang-like 2 (van gogh, Drosophila).

Is VANGL2 conserved across species?

Yes. The human VANGL2 gene has orthologues in mouse and rat, both named Vangl2, and the protein belongs to the Vang family, named after Drosophila van gogh. Check the datasheet of any product for its validated reactivity.
