Angiopoietin-1 Angiopoietin-1 · Ang1

Angiopoietin-1 (ANGPT1, also known as Ang1, AGPT-1 and KIAA0003) is a secreted glycoprotein of the angiopoietin family. It binds and activates the endothelial receptor TEK/TIE2, regulating angiogenesis, endothelial cell survival and the maintenance of vascular stability. Target information

What is Angiopoietin-1 (angiopoietin 1)?

Angiopoietin-1 binds and activates the TEK/TIE2 receptor by inducing its dimerisation and tyrosine phosphorylation. Through this receptor it regulates endothelial cell survival, proliferation, migration, adhesion and spreading, reorganisation of the actin cytoskeleton and the maintenance of vascular quiescence. Its effects depend on context. In quiescent vessels, where endothelial cells make tight contacts, ANGPT1 oligomers recruit TEK to cell-cell contacts and form complexes with TEK molecules on adjoining cells. This preferentially activates phosphatidylinositol 3-kinase and AKT1 signalling, inhibiting angiogenesis and promoting vascular stability.

In migrating endothelial cells that lack cell-cell adhesions, Ang1 instead recruits TEK to contacts with the extracellular matrix. This leads to focal adhesion complexes, activation of PTK2/FAK and the downstream kinases MAPK1/ERK2 and MAPK3/ERK1, and ultimately sprouting angiogenesis. The protein is required for normal angiogenesis and heart development during embryogenesis. It mediates blood vessel maturation and stability, inhibits endothelial permeability, and acts in endothelial developmental processes later than, and distinct from, those of VEGF. It also mediates reciprocal interactions between the endothelium and the surrounding matrix and mesenchyme.

The ANGPT1 gene encodes a secreted glycoprotein that belongs to the angiopoietin family, which is part of the fibrinogen C domain-containing group of receptor ligands. All angiopoietins bind with similar affinity to an endothelial cell-specific tyrosine-protein kinase receptor. Mutations in the gene are associated with hereditary angioedema. Mouse and rat orthologs are both designated Angpt1.

Angiogenesis research

Ang1 acts through TEK/TIE2 to activate or inhibit angiogenesis depending on context, making it central to studies of vessel growth.

Vascular stability and maturation

ANGPT1 promotes blood vessel maturation and stability and maintains vascular quiescence in vessels with tight endothelial cell contacts.

Endothelial permeability studies

Angiopoietin-1 inhibits endothelial permeability and mediates interactions between the endothelium and the surrounding matrix and mesenchyme.

Cardiovascular development

ANGPT1 is required for normal angiogenesis and heart development during embryogenesis, and its mutations are linked to hereditary angioedema.

Background

Angiopoietin-1 frequently asked questions

What does Angiopoietin-1 do?

Angiopoietin-1 binds TEK/TIE2 and induces its dimerisation and tyrosine phosphorylation. It supports endothelial cell survival, migration and adhesion, and promotes vessel maturation and stability. Depending on context, it either inhibits angiogenesis in quiescent vessels or stimulates sprouting angiogenesis in migrating endothelial cells.

What other names does ANGPT1 go by?

The protein is called Angiopoietin-1, and the gene symbol is ANGPT1 (HGNC name: angiopoietin 1). Recorded aliases include Ang1, AGPT-1 and KIAA0003. Researchers may therefore find Ang1 or AGPT-1 used in the literature for the same target.

Is Angiopoietin-1 conserved and linked to disease?

Orthologs exist in mouse and rat, both named Angpt1, and the gene is a member of the angiopoietin family. Mutations in human ANGPT1 are associated with hereditary angioedema. Check the datasheet of any product for its validated reactivity.
