VCAM-1 Vascular cell adhesion molecule 1 · CD106

VCAM-1 (vascular cell adhesion molecule 1, also known as CD106) is a cell surface sialoglycoprotein of the immunoglobulin superfamily, expressed mainly on cytokine-activated endothelium. It regulates leukocyte adhesion to the endothelium and contributes to immune surveillance and inflammation. Target information

What is VCAM-1 (vascular cell adhesion molecule 1)?

VCAM-1 is a cell adhesion glycoprotein predominantly expressed on the surface of endothelial cells, where it plays an important role in immune surveillance and inflammation. It acts as a major regulator of leukocyte adhesion to the endothelium through interaction with different types of integrins. During inflammatory responses, VCAM-1 binds ligands on the surface of activated endothelial cells to initiate the activation of calcium channels and the plasma membrane-associated small GTPase RAC1, leading to leukocyte transendothelial migration.

VCAM-1 also serves as a quality-control checkpoint for entry into bone marrow, providing a 'don't-eat-me' stamping in the context of major histocompatibility complex (MHC) class-I presentation. The protein is found at the cell membrane and in a secreted form.

The VCAM1 gene encodes a type I membrane protein that belongs to the Ig superfamily and is classed by HGNC among the CD molecules, receptor ligands and the Ig-like cell adhesion molecule family, including C2-set and I-set domain containing proteins. It is expressed by cytokine-activated endothelium and mediates leukocyte-endothelial cell adhesion and signal transduction. NCBI Gene notes that VCAM-1 may play a role in the development of atherosclerosis and rheumatoid arthritis, and describes three alternatively spliced transcripts encoding different isoforms. Mouse and rat orthologs are both designated Vcam1, which allows comparative work in standard rodent models.

Inflammation research

VCAM-1 is a key mediator of leukocyte adhesion to activated endothelium, making it central to studies of inflammatory responses.

Leukocyte transendothelial migration

VCAM-1 engagement activates calcium channels and RAC1, linking endothelial ligand binding to leukocyte migration across the vessel wall.

Atherosclerosis and arthritis

VCAM-1 may play a role in the development of atherosclerosis and rheumatoid arthritis, so it is studied in vascular and joint disease.

Immune surveillance and bone marrow

VCAM-1 acts as a quality-control checkpoint for bone marrow entry through a 'don't-eat-me' signal linked to MHC class-I presentation.

Background

VCAM-1 frequently asked questions

What does VCAM-1 do?

VCAM-1 is an endothelial cell adhesion glycoprotein that regulates leukocyte adhesion through interaction with integrins. During inflammation it triggers calcium channel and RAC1 activation, leading to leukocyte transendothelial migration. It also acts as a bone marrow entry checkpoint through a 'don't-eat-me' signal.

What other names and isoforms does VCAM-1 have?

VCAM-1 is encoded by the VCAM1 gene, and its alternative name is CD106. UniProt lists the protein as vascular cell adhesion protein 1. NCBI Gene describes three alternatively spliced transcripts that encode different isoforms, and the protein exists in membrane-bound and secreted forms.

Is VCAM-1 conserved in mouse and rat, and is it linked to disease?

Yes. Both mouse and rat have a Vcam1 ortholog; check the datasheet of any product for its validated reactivity. NCBI Gene reports that VCAM-1 may play a role in the development of atherosclerosis and rheumatoid arthritis.
