How a high sensitivity ELISA works
The same four-step sandwich workflow as a standard ELISA — the sensitivity gain comes from higher-affinity antibody pairs, an amplified detection step and standards calibrated across a lower concentration range.
Capture
Add sample and standards to the pre-coated plate. The immobilised capture antibody binds the analyte from serum, plasma, supernatant or lysate.
Detect
A biotinylated detection antibody binds a second epitope, forming the antibody–analyte–antibody sandwich that gives the assay its specificity.
Amplify
Streptavidin–HRP conjugate binds the detection antibody, delivering multiple enzyme molecules per analyte and amplifying low-abundance signal.
Read
TMB substrate develops colour in proportion to analyte concentration. Stop the reaction and read absorbance at 450 nm against the standard curve.

Frequently asked questions
What makes an ELISA “high sensitivity”?
A high sensitivity ELISA reaches a lower limit of detection than a standard kit for the same analyte — typically single-digit picograms per millilitre rather than tens or hundreds. That comes from three things: higher-affinity matched antibody pairs, an amplified detection step (biotin–streptavidin–HRP rather than a directly conjugated antibody), and a standard curve recalibrated across a lower concentration range so the bottom of the curve is properly resolved.
How low can these kits actually detect?
It depends on the analyte. The most sensitive kits in the range reach around 0.19 pg/mL, and many sit between 0.3 and 5 pg/mL. Each product page lists the kit’s own sensitivity and detection range — check those figures against the concentrations you expect in your matrix before ordering.
Which sample types are validated?
Most kits are validated in serum, plasma, cell culture supernatant, cell lysate, tissue homogenate and other biological fluids. Validated matrices are listed per kit, because a kit validated in serum will not necessarily perform identically in urine or CSF without a matrix check.
Should I use a high sensitivity ELISA or a standard one?
Use a high sensitivity kit when your expected analyte concentration is at or below the standard kit’s lower limit of quantification, when your sample volume is limited so you cannot concentrate the sample, or when you need to resolve small fold-changes near baseline. If your analyte is abundant, a standard kit gives you a wider upper range and fewer dilution steps.
Do you have kits for small sample volumes?
Yes — several kits in the range are supplied in a small-sample-volume format, designed for cases where only a few microlitres of serum or plasma are available, such as serial bleeds from mouse studies. These are listed alongside the standard-format kits in the catalogue.
What species are covered?

What precision should I expect?
Intra- and inter-assay coefficients of variation are held below 10%. Each batch is checked for lot-to-lot consistency so results remain comparable across timepoints and between studies, which matters when you are tracking small changes at the low end of the curve.
