ELISpot kits for single-cell cytokine detection
Count the individual cells that secrete IFN-gamma, granzyme B, perforin, interleukins or TNF-alpha. Pre-coated ELISpot kits detect secretion at the single-cell level in conditions close to the in vivo environment, with minimal cell handling, for T-cell, NK-cell and vaccine response studies. ELISpot antibody pairs are available for labs that coat their own plates.

What's in an ELISpot kit
Pre-coated ELISpot kits include PVDF plates pre-coated with capture antibody, a detection antibody, alkaline phosphatase conjugate, BSA and ready-to-use BCIP/NBT substrate. You add your cells and stimulus.
ELISpot or ELISA?
An ELISA measures the total amount of a cytokine in a supernatant. ELISpot counts how many cells secrete it, which makes it far more sensitive for rare antigen-specific T cells and a standard read-out in vaccine and immunotherapy research.
Running the assay
Stimulate cells directly in the pre-coated wells, then develop the plate so that each secreting cell leaves one coloured spot. Count spots with an ELISpot reader or a dissecting microscope. Incubation times are on each datasheet.
