Choosing a primary antibody
Check three things on the datasheet: the application the antibody is validated for (WB, IHC, IF, flow or ELISA), the reactivity for your species, and the clonality. Monoclonal and rabbit monoclonal antibodies give lot-to-lot consistency; polyclonal antibodies recognise several epitopes and often give a stronger signal.
Secondary antibodies and conjugates
Pick a secondary antibody raised against the host species of your primary, with a label that suits your read-out: HRP for Western blot and ELISA, or fluorophores such as FITC, PE and APC for flow cytometry and imaging. Our guide on how to choose a secondary antibody walks through each step.
Validation you can check
Knockout-validated antibodies show loss of signal in a knockout cell line, the strongest evidence of specificity. Loading controls confirm equal loading on a blot, and ChIP antibodies are tested for chromatin immunoprecipitation.
