How Flow Cytometry Antibodies Work
Flow cytometry antibodies bind to specific antigens on the surface of cells or within cells, either directly conjugated to a fluorescent probe or paired with a fluorophore-conjugated secondary antibody. As cells pass single-file through a laser beam, forward scatter (FSC) reveals cell size and side scatter (SSC) reveals granularity, while the fluorescence signal from bound antibodies reveals marker expression.
- Immunophenotyping and cell sorting by fluorescence
- Cell cycle and proliferation analysis
- Viability analysis via dye exclusion
- Monoclonal antibodies give the lowest background for multi-color panels
Frequently Asked Questions
What is flow cytometry?
Flow cytometry is a technique used to analyze the physical and chemical properties of a population of cells or particles, including intracellular proteins, peptides and DNA, using laser-beam technology to assess size, shape and properties of individual cells.
How does flow cytometry work?
A cell suspension is fed into the cytometer where hydrodynamic focusing forces cells single-file through a laser beam; deflected and emitted light is captured by detectors to reveal cell size, complexity and marker expression.
Monoclonal or polyclonal antibodies for flow cytometry?
Monoclonal antibodies bind a single epitope and give the lowest background, making them the default choice for multi-color panels, while polyclonal antibodies can be a cost-effective option for single-marker detection.
How do I choose fluorophores for a multi-color panel?
Match brighter dyes to lower-expressing targets, check your cytometer's laser and filter configuration, and avoid combining dyes with heavily overlapping emission spectra in the same panel — use the channel chooser above as a starting point.
