Neutralising or receptor-blocking antibodies are a core in vivo tool for dissecting cytokine function — removing one signal at a time to test its role in immunity, inflammation, autoimmunity and the tumour microenvironment. This network groups the major axes by outcome.
Pro-inflammatory: IFN-γ and type I IFN drive STAT1 (Th1 / M1, MHC upregulation); TNF-α and IL-1 drive NF-κB acute inflammation; GM-CSF fuels myeloid inflammation. Th2 / allergy: IL-4 → STAT6 (IgE, eosinophilia, fibrosis). Immunosuppressive: IL-10 → STAT3 and TGF-β → SMAD3 (Treg induction, tolerance). T-cell growth: IL-2 → STAT5 (proliferation and survival).
